FDA approves first CRISPR therapy—here’s how it works against sickle cell

The Food and Drug Administration on Friday approved two gene therapies to treat sickle cell disease, one of the which is the first CRISPR/Cas9-based treatment to win regulatory approval in the US.

The announcement is a landmark in the treatment of sickle cell disease, a devastating condition in which red blood cells deform into a sickle shape and clog up blood vessels. Sickle cell disease affects around 100,000 people in the US, most commonly African Americans. It leads to anemia, vaso-occlusive events and crises (painful episodes in which small blockages starve tissue of oxygen), strokes, progressive and irreversible organ damage, decreased quality of life, and early death.

Until today, treatments have been limited. A bone marrow transplant from a genetically matched sibling can cure the condition more than 90 percent of the time, but only around 20 percent of people with the disease have such a genetically matched sibling donor. There are also multiple drugs available and supportive care, but these mainly reduce the severity of the disease. The new gene therapy treatments, on the other hand, have shown to be highly effective at preventing vaso-occlusive events and crises.

Beeper Mini, the Android app born from a reverse-engineering of Apple’s iMessage service, is currently broken, and it is unknown whether it will resume functioning.

Beeper desktop users received a message from co-founder Eric Migicovsky late on Friday afternoon, noting an “iMessage outage” and that “messages are failing to send and receive.” Reports had started piling up on Reddit around 2:30 pm Eastern. As of 5:30 pm, the app was still reporting errors in sending and receiving messages, with “Failed to lookup on sever: lookup request timed out.” Comments on Beeper’s status post on X (formerly Twitter) suggested mixed results, at best, among users.

The Verge, messaging with Migicovsky, reported that he “did not deny that Apple has successfully blocked Beeper Mini”; to TechCrunch, Migicovsky more clearly stated about an Apple cut-off: “Yes, all data indicates that.” To both outlets, Migicovsky offered the same comment, re-iterating his belief that it was in the best interests of Apple to let iPhone owners and Android users send encrypted messages to one another. Earlier this month, Microsoft posted a new entry in its list of known issues with fully up-to-date Windows 11 PCs: The HP Smart printer app was installing automatically on Windows 10 and Windows 11 PCs whether they had an HP printer installed or not, changing the names and icons of their connected printers and causing error messages.

Affected PCs will usually appear to have an HP LaserJet M101-M106 connected, so look for that model number in your list of printers (people who actually own one of those HP LaserJets presumably won’t have problems). All versions of Windows 11 are affected, plus all currently supported versions of Windows 10; Windows Server versions going back to 2012 can also be affected.

Microsoft continues to look into the issue, but in an update posted yesterday, the company stated unambiguously that HP was not to blame. The company also says that most printers should continue to work fine, and that they “will continue to use the expected drivers for printer operations.” But if your printer relies on a third-party app for additional functionality, that may be broken.

According to a report in Bloomberg, Tang Tan, vice president of Product Design, is leaving Apple, and his departure heralds a shuffle of executives heading up some of the company’s most important products.

Sometimes, you might wonder just how much a specific executive influences the grand scheme of things, but the report claims that people within Apple see Tan’s departure as “a blow,” clarifying that he “made critical decisions about Apple’s most important products.” His team reportedly had “tight control” over the look and functionality of those products.

Tan oversaw major aspects of iPhone and Apple Watch design, and he was the executive overseeing accessories and AirPods, as well. He reported to John Ternus, Apple’s senior vice president of Hardware Engineering, who is likely a more widely known name.

Stealthy and multifunctional Linux malware that has been infecting telecommunications companies went largely unnoticed for two years until being documented for the first time by researchers on Thursday.

Researchers from security firm Group-IB have named the remote access trojan “Krasue,” after a nocturnal spirit depicted in Southeast Asian folklore “floating in mid-air, with no torso, just her intestines hanging from below her chin.” The researchers chose the name because evidence to date shows it almost exclusively targets victims in Thailand and “poses a severe risk to critical systems and sensitive data given that it is able to grant attackers remote access to the targeted network.

Some do it horizontally, some do it vertically, some do it sexually, and some asexually. Then there are some organisms that would rather grow a butt that develops into an autonomous appendage equipped with its own antennae, eyes, and brain. This appendage will detach from the main body and swim away, carrying gonads that will merge with those from other disembodied rear ends and give rise to a new generation.

Megasyllis nipponica really exists on Earth. Otherwise known as the Japanese green syllid worm, it reproduces by a process known as stolonization, which sounds like the brainchild of a sci-fi horror genius but evolved in some annelid (segmented) worms to give future generations the best chance at survival. What was still a mystery (until now) was exactly how that bizarre appendage, or stolon, could form its own head in the middle of the worm’s body. Turns out this is a wonder of gene regulation.

Back in April, we ran a series of useful and/or somewhat goofy prompts through Google’s (then-new) PaLM-powered Bard chatbot and OpenAI’s (slightly older) ChatGPT-4 to see which AI chatbot reigned supreme. At the time, we gave the edge to ChatGPT on five of seven trials, while noting that “it’s still early days in the generative AI business.”

Now, the AI days are a bit less “early,” and this week’s launch of a new version of Bard powered by Google’s new Gemini language model seemed like a good excuse to revisit that chatbot battle with the same set of carefully designed prompts. That’s especially true since Google’s promotional materials emphasize that Gemini Ultra beats GPT-4 in “30 of the 32 widely used academic benchmarks” (though the more limited “Gemini Pro” currently powering Bard fares significantly worse in those not-completely-foolproof benchmark tests).

This time around, we decided to compare the new Gemini-powered Bard to both ChatGPT-3.5—for an apples-to-apples comparison of both companies’ current “free” AI assistant products—and ChatGPT-4 Turbo—for a look at OpenAI’s current “top of the line” waitlisted paid subscription product (Google’s top-level “Gemini Ultra” model won’t be publicly available until next year). We also looked at the April results generated by the pre-Gemini Bard model to gauge how much progress Google’s efforts have made in recent months.

Google Podcasts has been sitting on Google’s death row for a few months now since the September announcement. Now, a new support article details Google’s plans to kill the product, with a shutdown coming in April 2024.

Google Podcasts (2016–2024) is Google’s third attempt at a podcasting app after the Google Reader-powered Google Listen (2009–2012) and Google Play Music Podcasts (2016–2020). The product is being shut down in favor of podcast app No. 4, YouTube Podcasts, which launched in 2022.

Google support article details how you can take your subscriptions with you. If you want to move from Google Podcasts to YouTube Podcasts, Google makes that pretty easy with a one-click button at music.youtube.com/transfer_podcasts. If you want to leave the Google ecosystem for something with less of a chance of being shut down in three to four years, you can also export your Google Podcast subscriptions as an OPML file at podcasts.google.com/settings. Google says exports will be available until August 2024.

Modern EVs have some pretty huge batteries, but like their gas-powered counterparts, the main thing they do is sit in one place, unused. The Ford F-150 Lightning was built with two-way power in mind, and soon it might have a use outside emergency scenarios.

Ford and Resideo, a Honeywell Home thermostat brand, recently announced the EV-Home Power Partnership. It’s still in the testing phases, but it could help make EVs a more optimal purchase. Put simply, you could charge your EV when it’s cheap, and when temperatures or demand make grid power time-of-use expensive (or pulled from less renewable sources), you could use your truck’s battery to power the AC. That would also help with grid reliability, should enough people implement such a backup.

The F-150 Lightning already offers a whole-home backup power option, one that requires the professional installation of an 80-amp Ford Charge Station Pro and a home transfer switch to prevent problems when the grid switches back on. Having a smart thermostat allows for grid demand response, so the F-150 would be able to more actively use its vehicle-to-home (V2H) abilities.

Elon Musk yesterday appealed to the Supreme Court in a last-ditch effort to terminate his settlement with the Securities and Exchange Commission. Musk has claimed he was coerced into the deal with the SEC and that it violates his free speech rights, but the settlement has been upheld by every court that’s reviewed it so far.

In his petition asking the Supreme Court to hear the case, Musk said the SEC settlement forced him to “waive his First Amendment rights to speak on matters ranging far beyond the charged violations.”

The SEC case began after Musk’s August 2018 tweets stating, “Am considering taking Tesla private at $420. Funding secured” and “Investor support is confirmed. Only reason why this is not certain is that it’s contingent on a shareholder vote.” The SEC sued Musk and Tesla, saying the tweets were false and “led to significant market disruption.”

Marbled paper is an art form that dates back at least to the 17th century, when European travelers to the Middle East brought back samples and bound them into albums. Its visually striking patterns arise from the complex hydrodynamics of paint interacting with water, inspiring a winning video entry in this year’s Gallery of Fluid Motion.

The American Physical Society’s Division of Fluid Dynamics sponsors the gallery each year as part of its annual meeting, featuring videos and posters submitted by scientists from all over the world. The objective is to highlight “the captivating science and often breathtaking beauty of fluid motion” and to “celebrate and appreciate the remarkable fluid dynamics phenomena unveiled by researchers and physicists.”

The three videos featured here are the winners of the Milton Van Dyke Awards, which also included three winning posters. There were three additional general video winners—on the atomization of impinging jets, the emergent collective motion of condensate droplets, and the swimming motion of a robotic eel—as well as three poster winners. Google is facing controversy among AI experts for a deceptive Gemini promotional video released Wednesday that appears to show its new AI model recognizing visual cues and interacting vocally with a person in real time. As reported by Parmy Olson for Bloomberg, Google has admitted that was not the case. Instead, the researchers fed still images to the model and edited together successful responses, partially misrepresenting the model’s capabilities.

“We created the demo by capturing footage in order to test Gemini’s capabilities on a wide range of challenges,” a spokesperson said. “Then we prompted Gemini using still image frames from the footage, & prompting via text,” a Google spokesperson told Olson. As Olson points out, Google filmed a pair of human hands doing activities, then showed still images to Gemini Ultra, one by one. Google researchers interacted with the model through text, not voice, then picked the best interactions and edited them together with voice synthesis to make the video.

Right now, running still images and text through massive large language models is computationally intensive, which makes real-time video interpretation largely impractical. That was one of the clues that first led AI experts to believe the video was misleading.

On November 22, a few days after OpenAI fired (and then re-hired) CEO Sam Altman, The Information reported that OpenAI had made a technical breakthrough that would allow it to “develop far more powerful artificial intelligence models.” Dubbed Q* (and pronounced “Q star”) the new model was “able to solve math problems that it hadn’t seen before.”

Reuters published a similar story, but details were vague.

Both outlets linked this supposed breakthrough to the board’s decision to fire Altman. Reuters reported that several OpenAI staffers sent the board a letter “warning of a powerful artificial intelligence discovery that they said could threaten humanity.” However, “Reuters was unable to review a copy of the letter,” and subsequent reporting hasn’t connected Altman’s firing to concerns over Q*.

Back when WarCraft, StarCraft, and other real-time strategy games were all the rage, I could never actually play them against other people. Even playing against the computer, I might only eke out a victory through dumb luck or an opponent’s huge mistake.

The problem was, I was never ready to attack until I had my base perfectly in order—until the workers carrying oil or crystals or whatnot had the most efficient route from the mine to the base or until my buildings were arranged for optimal use of the revealed land. I just needed one more little guy, one more tower. OK, maybe two. I’m a turtler’s turtler.

Against the Storm, which releases from Early Access on Steam for Windows today, has been a deeply satisfying outlet for this pent-up need to build and prosper—in a delightfully WarCraft-ian manner—without the messy business of war. There is still adversity: an ever-advancing “Impatience” meter, hostile spirits you uncover in the woods, and the typical constraints of resources, supply chains, and worker morale. Plus the rainstorms in the title, which occur both in-level, slowing you down, and at a macro level, washing away your little towns to make you start again.

Welcome to Edition 6.22 of the Rocket Report! We’re nearing the end of 2023, and it’s been an incredible year for rocket debuts. Early in the year we saw small lift vehicles from Relativity Space and ABL, and in the spring Japan’s H3 and SpaceX’s Starship rocket. There’s one big one left: United Launch Alliance’s Vulcan booster. That will be a nice stocking stuffer to end the year on Christmas Eve.

Vega has a missing parts problem. In unhappy news for Italian rocket-maker Avio, two of the four propellant tanks on the fourth stage of the Vega rocket—the upper stage, which is powered by dimethylhydrazine and nitrogen tetroxide fuel—went missing earlier this year. Now, it seems that the propellant tanks have been found. However, Ars reports, the tanks were recovered in a dismal state, crushed alongside metal scraps in a landfill. This is a rather big problem for Avio, as this was to be the final Vega rocket launched, and the production lines are now closed for this hardware.

Fungi join the list of organisms that can control when ice forms

While it may be the reason behind tires skidding, pipes bursting, and closed roads making traffic a nightmare, ice doesn’t always form as easily as it seems. It often gets an assist from proteins made by fungi. 

Never mind the common thinking that ice forms at 0° C (32° F). Though this is water’s freezing point, pure water will only freeze when temperatures plummet as low as minus 46° C (minus 50.8° F). So why does it usually freeze at zero anyway? Organisms such as bacteria, insects, and fungi produce proteins known as ice nucleators (non-protein nucleators can also be of abiotic origin). These proteins can kick-start the formation, or nucleation, of ice at higher temperatures than pure water would freeze at.

While the exact reason fungi make these proteins remains unknown, researchers Valeria Molinero of the University of Utah and Konrad Meister of Boise State University led a study that has revealed more about how fungal ice nucleators can both promote and hold back ice formation more efficiently than those of many other life-forms.

Google is dealing with its second “lost data” fiasco in the past few months. This time, it’s Google Drive, which has been mysteriously losing files for some people. Google acknowledged the issue on November 27, and a week later, it posted what it called a fix.

It doesn’t feel like Google is describing this issue correctly; the company still calls it a “syncing issue” with the Drive desktop app versions 84.0.0.0 through 84.0.4.0. Syncing problems would only mean files don’t make it to or from the cloud, and that doesn’t explain why people are completely losing files. In the most popular issue thread on the Google Drive Community forums, several users describe spreadsheets and documents going missing, which all would have been created and saved in the web interface, not the desktop app, and it’s hard to see how the desktop app could affect that. Many users peg “May 2023” as the time documents stopped saving. Some say they’ve never used the desktop app.

Drive has multiple ways of syncing files, which complicates any explanation or diagnosis of what’s going on. The most suspect mode in the whole fiasco is the default “file streaming,” mode which is actually cloud-first. Files get removed from your computer and stored in the cloud, saving space and leaving only a web link in their place. Perhaps a “syncing” issue could remove files from your computer before uploading (that still doesn’t explain the claims of missing web documents, though).
